BIOENTAL is a Singapore-based life science distributor specialising in bacterial endotoxin testing solutions, BET consumables, LAL and recombinant endotoxin testing technologies, method validation support and laboratory troubleshooting for pharmaceutical, biotechnology, medical device, healthcare and research laboratories across Singapore and Southeast Asia.

What Is Endotoxin Testing?

Endotoxin testing is a quality control process used to detect bacterial endotoxins, which are components found in the outer membrane of Gram-negative bacteria. Endotoxins may cause fever, inflammation or serious adverse reactions if present in pharmaceutical products, medical devices or healthcare-related products.

Bacterial Endotoxin Testing, commonly known as BET, is therefore an important test for laboratories involved in pharmaceutical manufacturing, medical device production, biologics, healthcare products and research applications.

Why BET Matters

BET helps laboratories assess whether products or materials meet acceptable endotoxin limits. It is especially important for products that come into contact with blood, cerebrospinal fluid, injectable drugs or other sterile applications.

Reliable BET results depend not only on reagents, but also on sample preparation, consumables, water quality, analyst technique, equipment and workflow control.

Common BET Methods

Common endotoxin testing methods include:

  • Gel clot testing
  • Kinetic chromogenic testing
  • Kinetic turbidimetric testing
  • Recombinant Factor C testing
  • Recombinant cascade reagent testing

Each method has different strengths. The most suitable method depends on the sample type, regulatory expectations, sensitivity requirements, laboratory workflow and validation outcome.

LAL-Based Endotoxin Testing

LAL-based endotoxin testing uses reagents derived from horseshoe crab blood. Common LAL methods include gel clot, chromogenic and turbidimetric methods. These methods are widely used in pharmaceutical and medical device laboratories.

Recombinant Endotoxin Testing

Recombinant endotoxin testing technologies, including rFC and rCR, are animal-free alternatives that may support laboratories seeking sustainable and modern endotoxin testing options.

Laboratories considering recombinant methods should assess method suitability, validation requirements and acceptance within their intended regulatory or quality framework.

BET Validation

Before routine testing, laboratories must show that the selected BET method is suitable for the sample. This may include checking for inhibition, enhancement, interference and acceptable recovery.

Common validation challenges include:

  • Failed positive product control (PPC)
  • Low endotoxin recovery
  • Sample interference
  • Poor dilution strategy
  • Contamination from consumables
  • Inconsistent analyst technique

BET Troubleshooting

When BET results are inconsistent, laboratories should review possible causes such as:

  • Reagent preparation
  • Endotoxin standard handling
  • Sample dilution
  • Consumables
  • Water quality
  • Equipment condition
  • Environmental contamination
  • Analyst workflow

A systematic troubleshooting process helps reduce repeat testing and improves confidence in test results.

Endotoxin-Free Consumables

Consumables play an important role in BET reliability. Tubes, pipette tips, plates and other materials should be suitable for endotoxin testing to reduce contamination risk and workflow variability.

BIOENTAL supports laboratories with endotoxin-free consumables and practical workflow guidance.
